A stabilised emulsion of egg yolk for use in culture media. It may be added directly to nutrient media for the identification of Clostridium, Bacillus and Staphylococcus species by their lipase activity.
Egg Yolk Emulsion, Oxoid Directions:
Suspend 21.5g of MYP Agar (CM0929) in 450ml of distilled water and bring gently to the boil to
dissolve. Sterilize by autoclaving at 121°C for 15 minutes. Cool to approximately 50°C and
aseptically add 50ml of Egg Yolk Emulsion (SR0047C) and the contents of one vial of Polymyxin B
Supplement (SR0099E) reconstituted as directed. Mix well and pour into sterile Petri dishes.
Alternatively, suspend 8g of Blood Agar Base (CM0055) in 200ml of distilled water and add 2g of
sodium chloride. Sterilize by autoclaving at 121°C for 15 minutes. Cool to approximately 50°C and
aseptically add 20ml Egg Yolk Emulsion (SR0047C). Mix well and pour into sterile Petri dishes.

Keep at temperatures between 2° and 8 °C. Use before expiry date on the label.
Precautions
On this medium Bacillus cereus is indistinguishable from Bacillus thuringiensis.
Identify Bacillus cereus by colony form, colour, egg yolk hydrolysis and confirm with cell and spore morphology.
Occasional strains of Bacillus cereus show weak or negative egg yolk reactions.

Agar Preparation:
To Prepare the Agar Base:
Suspend 23g in 500ml of distilled water and heat gently until the agar is completely dissolved. Sterilise by autoclaving at 121°C for 10 minutes. Allow the medium to cool to 50°C.
To Prepare Tryptose Sulphite Cycloserine Agar (TSC Agar):
To 500ml of Agar base cooled to 50°C add the rehydrated contents of 1 vial of TSC supplement (SR0088) and 25ml of egg yolk emulsion (SR0047). Mix well and pour into sterile Petri dishes.
To Prepare Egg Yolk Free TSC Agar:
To 500 ml of Agar base cooled to 50°C add the rehydrated contents of 1 vial of TSC supplement (SR0088). Mix well and pour into sterile Petri dishes.
To Prepare Shahidi-Ferguson Perfringens Agar (SFP Agar):
To 500 ml of Agar base cooled to 50°C add the rehydrated contents of 1 vial of SFP supplement (SR0093) and 25ml of egg yolk emulsion (SR0047) mix well and pour into sterile Petri dishes.
To Prepare Agar for an Overlay:
For TSC or SFP Agar used as an overlay, the egg yolk emulsion (SR0047) is omitted. Its inclusion does not improve the lecithinase reaction and diminishes the visibility of the colonies.

Suspend 63g of Baird-Parker Agar (ISO) Base in 1,000ml of distilled water. Boil to dissolve the medium and sterilise by autoclaving at 121°C for 15 minutes. Cool to 47°C and aseptically add 50ml of Egg Yolk Tellurite Emulsion (SR0054). Mix well and pour into sterile Petri dishes.

Precautions
Atypical colonies have the same size as typical colonies but can be shining black with or without a narrow white edge; the clear zone or opacity halo can also be absent or barely visible. Grey colonies, free of clear zone, can also be regarded as atypical.
Regard all typical and atypical colonies as coagulase-positive staphylococci irrespective of negative reactions on the medium and carry out further tests.
Colonies of some contaminating organisms growing in close proximity to the coagulase positive colonies may partially digest the coagulase halo reaction.

Preston Campylobacter Selective Enrichment Broth, Oxoid Preparation:
Dissolve 12.5g of Nutrient Broth No.2 (CM0067) in 475ml of distilled water and sterilise by autoclaving at 121°C for 15 minutes. Cool to 50°C or below. Aseptically add 25ml of Lysed Horse Blood (SR0048), 1 vial of Preston Campylobacter Selective Supplement (SR0117 or SR0204) and 1 vial of Campylobacter Growth Supplement (SR0232). Precautions
This medium should not be used if agents causing systemic mycoses are being sought9. If such agents are suspected e.g. cryptococcus, histoplasma etc., either Dermasel Agar Base without antibiotic supplement must be used in parallel or Brain Heart Infusion Agar CM0375.
If the fungal agent sought is suspected to be nutritionally fastidious, the use of Brain Heart Infusion Agar is particularly helpful.
Dermasel Selective Supplement SR0075 contains a toxic concentration of cycloheximide. Note the precautions to be taken under HAZARDS.

SeaKem® LE Agarose was the first and is still the world’s most trusted agarose for nucleic acid electrophoresis. Ideal for routine analysis of nucleic acids by gel electrophoresis and blotting, each SeaKem® LE Gel sharply resolves DNA and provides consistent resolution from lot-to-lot. This molecular biology grade agarose has no detectable DNase or RNase activity and forms strong gels with low background upon ethidium bromide, SYBR® Green, or GelStarTM Staining. Due to its low EEO, DNA will have a high electrophoretic mobility. SeaKem® LE Agarose is also recommended for protein gel electrophoresis in applications such as Ouchterlony and radial immunodiffusion (RID).
