Vogel-Johnson Agar, Oxoid Preparation:
Suspend 61g in 1 litre of distilled water and bring gently to the boil to dissolve completely. Sterilise by autoclaving at 121°C for 15 minutes. Cool to 50°C and add 5.7ml of sterile 3.5% potassium tellurite solution SR0030 (equivalent to 20ml of 1% potassium tellurite).

Store the dehydrated medium at 10-30°C and use before the expiry date on the label.
Store the prepared medium at 2-8°C.
Precautions
All presumptive Staphylococcus aureus colonies should be confirmed with further tests. Do not heat the medium after the addition of potassium tellurite.

Application
Cytochrome c has been identified as an important mediator in apoptotic pathways. The release of mitochondrial cytochrome c into the cytoplasm stimulates apoptosis and is commonly used as an indicator of the apoptotic process in the cell. Investigation on the effect of Paris Saponin I (PS I) on human gastric carcinoma cell growth (SGC7901 cells) have shown an elevated level cytoplasmic cytochrome c. Results are inhibition of proliferation in SGC7901 cells by inducing mitochondria-dependent apoptosis through cytochrome c.
Biochem/physiol Actions
The ready interconversion of cytochrome c between ferrous and ferric states makes it an efficient biological electron carrier. It plays a vital role in cellular oxidations in both plants and animals. Suspend 27g of Brilliance Salmonella Agar Base in 500ml of distilled water. Add the contents of 1 vial of Salmonella Selective Supplement, resuspended as directed. Mix well and sterilise by bringing to the boil with frequent agitation. Cool to around 50ºC, mix well and pour into sterile Petri dishes.
NOTE: It is critical that the selective supplement is added prior to heating. When preparing volumes greater than 1 litre contact Oxoid Technical Support for directions.

Salmonella Selective Supplement should be stored in the dark at 2-8ºC. When stored as directed, the un-opened products will remain stable until the expiry date printed on the packaging.

Q5® High-Fidelity DNA Polymerase sets a new standard for both fidelity and robust performance. With the highest fidelity amplification available (~280 times higher than Taq), Q5 DNA Polymerase results in ultra-low error rates. Q5 DNA Polymerase is composed of a novel polymerase that is fused to the processivity-enhancing Sso7d DNA binding domain, improving speed, fidelity and reliability of performance.
