YeastBuster™ Protein Extraction Reagent is formulated for a fast, efficient and gentle extraction of active proteins from yeast cells. The reagent avoids harsh conditions of vigorous mechanical or chemical treatment that often result in heat degradation of target proteins. The proprietary formulation utilizes a mix of mild detergent, protein stabilization buffer, and tris(hydroxypropyl) phosphine (THP) reducing agent (THP concentrate provided separately). This powerful combination eliminates the inconsistencies associated with tedious mechanical disruption of yeast cells with glass bead abrasives, ultrasonication and pressure disruption, or enzymatic digestion with β-1,3-glucanase lytic enzymes. The reagent has been tested with Saccharomyces cerevisiae, Pichia pastoris, P. stipidis, and Schizosaccharomyces pombe strains and with plant cells.

In practice, cells are harvested by centrifugation and suspended in YeastBuster. Following a brief incubation, insoluble cell debris is removed by centrifugation, and the clarified extract is ready to use. In addition to greater total protein yields in crude extracts and recovery of enzymatically active protein, the extracts are fully compatible with GST•Bind™ and Ni-NTA His•Bind® immobilized metal affinity chromatography (IMAC) purification methods. The addition of 2.5% (v/v) Egg Yolk Emulsion (SR0047) to the medium may aid the recovery of damaged Listeria.

Add 38g of Mueller Hinton Agar to 1 litre of distilled water. Bring to the boil to dissolve the medium completely. Sterilize by autoclaving at 121°C for 15 minutes.

Precautions
Incubation in a carbon dioxide enriched atmosphere is not recommended because of its pH effect on the medium. If it is imperative to use CO2 then known control organisms should be included with the test plates to measure its effect.
Carbohydrates should not be added to Mueller-Hinton Agar because they may influence the rate of growth of the organism and the resulting pH of the medium.
The addition of lysed horse blood to the medium may further reduce the levels of thymidine and prevent the growth of thymidine-dependent organisms.

The medium may not support the growth of some fastidious strains for example microorganisms that require thymine and thymidine for growth. Close adherence to the methodology is required to obtain satisfactory results.

MYP Agar 500g

Selectively enumerate Bacillus cereus in food samples with Thermo Scientific™ Oxoid™ MYP Agar (Dehydrated). The medium relies upon mannitol fermentation and phospholipase C production to differentiate Bacillus cereus from other members of the species.

Mannitol-negative Bacillus cereus form pink colored colonies due to the presence of Phenol Red Indicator. Egg yolk emulsion helps in the differentiation of phospholipase C producing colonies, which are surrounded by a zone of the white precipitate. Polymixin B Sulphate acts as the inhibitor to restrict the growth of Gram-negative bacteria.

Burkholderia Cepacia Agar Base 500g

The slower growing B. cepacia can be missed on conventional media, such as blood or MacConkey Agar, due to overgrowth caused by other faster growing organisms found in the respiratory tract of CF patients, such as mucoid Klebsiella species,

Buffer tablets pH 7.2 – for preparing buffer solution acc. to WEISE for staining of blood smears, are used to prepare buffer solutions according to WEISE and pH-buffered staining solutions. Phosphate buffer solutions are used in hematology, for the dilution of staining solutions and for the rinsing steps in the staining process. The buffer tablets acc. to Weise will produce a buffered solution (pH6.4/pH6.8/pH7.2) that stabilizes the staining result and gives reproducible staining results. Buffer solution acc. to Weise pH 6.4 results in bright-orange stained erythrocytes and differentiated stained cells with nuclei, whereby the erythrocytes are stained orange when using pH 6.8. Buffer solution acc. to Weise pH 7.2 produces grayish stained erythrocytes and intense red-violet stained cells with nuclei.

The MQuant® pH tests are suited for all media in environmental analysis and in industrial in-process controls. Our special method of manufacture gives the pH indicator strips decisive advantages over the indicator paper: since the indicator does not bleed, the strip can be left in the measurement medium without contaminating it. Using these test strips is fairly easy, the pH value is measured semiquantitatively by visual comparison of the reaction zone on the test strip with the fields of the color scale, perfectly matching the reaction color. The clever Safetyedge boxes allow for safe storage and convenient withdrawal of the test strips. SeaKem® LE Agarose was the first and is still the world’s most trusted agarose for nucleic acid electrophoresis.
