The Giemsa’s Azure Eosin Methylene Blue solution can be used for the staining of blood and bone marrow smears, paraffin sections and clinical-cytological specimens. Lysine Medium, Oxoid Preparation:
Suspend 6.6g in 100ml distilled water containing 1.0ml Potassium lactate SR0037. Bring to the boil to dissolve completely. Agitate frequently to prevent superheating. Cool to 50°C and add 0.1ml of lactic acid 10% SR0021 to adjust to pH 4.8 ± 0.2. Dispense into Petri dishes and remove surface moisture by drying at 37°C.

Suspend 21.5g of Wilkins-Chalgren Anaerobe Agar (CM0619) in 475ml of distilled water containing 0.5ml `Tween 80’. Bring to the boil to dissolve completely and sterilise by autoclaving at 121°C for 15 minutes. Cool to 50-55°C and aseptically add the contents of 1 vial of N-S Anaerobe Supplement (SR0107) rehydrated as directed, together with 25ml of defibrinated blood (SR0050/SR0051). Mix gently and pour into sterile Petri dishes.

Modified ISO Preparation:
