Hoyle Medium Base, Oxoid Composition

Hoyle Medium, Oxoid Preparation:
Suspend 40g in 1 litre of distilled water and bring to the boil to dissolve completely. Sterilise by autoclaving at 121°C for 15 minutes. Cool to 55°C and add 50ml of Laked Horse Blood SR0048 and 10ml of 3.5% Potassium Tellurite solution SR0030, mix, and pour plates.

Storage conditions and Shelf life
Store the dehydrated medium at 10-30°C and use before the expiry date on the label.
Store the prepared plates at 2-8°C.

Precautions
It should be noted that not all corynebacteria produce the typical colonies described above – so in all cases it is advisable to use Hoyle medium in conjunction with the additional media and tests mentioned above.

Selectively isolate Neisseria species with Thermo Scientific™ Oxoid™ GC Agar Base (Dehydrated). GC Agar Base has been formulated to include Special Peptone LP0072 which is a mixture of meat and plant enzymatic digests. The presence of starch in the medium ensures that toxic metabolites produced by Neisseria are absorbed. Phosphate buffers are included to prevent changes in pH due to amine production that would affect the survival of the organism

ONE Broth Salmonella Selective Supplement, Oxoid Composition

ONE Broth Salmonella Selective Medium Preparation:
Suspend 5.6g of ONE Broth-Salmonella Base in 225ml of distilled water. Sterilize by autoclaving at 121°C for 15 minutes. Cool to below 50°C and add the contents of 1 vial of ONE Broth-Salmonella Supplement (SR0242), resuspended as directed.

Storage conditions and Shelf life
The ONE Broth-Salmonella Selective Supplement should be stored in the dark at 2-8ºC. When stored as directed, the unopened products will remain stable until the expiry date printed on the packaging.

Once prepared, ONE Broth-Salmonella can be stored for up to 2 weeks when made from CM1091 and SR0242 according to the manufacturer’s instructions and stored at 2-8ºC, in the dark. A longer shelf life may be attainable, but should be validated under the relevant, local manufacturing and storage conditions.

Enrich and recover Vibrio species from food and water samples with Thermo Scientific™ Oxoid™ Alkaline Saline Peptone Water (ISO) (Dehydrated), while adhering to ISO/TS 21872-1&2:2007. This medium is selective by exploiting the ability of Vibrio species to tolerate alkaline conditions and high sodium chloride levels. The peptone content provides the conditions for growth and recovery of Vibrio species, supplying vitamins, minerals, and nitrogen in the form of amino acids.

Alkaline Saline Peptone Water (ISO), Oxoid Preparation:

Dissolve 40g in 1 litre of distilled water. Mix well and distribute into final containers. Sterilize by autoclaving at 121°C for 15 minutes.

Enumerate and differentiate bacteria in dairy products with this standard Thermo Scientific™ Oxoid™ China Blue Lactose Agar (Dehydrated). China Blue Lactose Agar, originally formulated by Brandl and Sobeck-skal, is a standard non-inhibitory solid medium used for the differentiation of lactose fermenters from non-lactose fermenters in milk.

The china blue in the medium serves as a pH indicator to differentiate lactose fermenters from non-lactose fermenters. The medium does not contain any inhibitory substances therefore all the organisms present in milk sample grow luxuriantly on this medium.

Suspend 35g in 1 litre of distilled water. Boil to dissolve completely. Sterilise by autoclaving at 121°C for 15 minutes.

Precautions
It is important to remember that Gram positive and negative cocci and bacilli can grow on this medium. Always confirm the organism morphology and Gram reaction.

Description

LB Agar Miller from Merck is a commonly used medium in microbiology laboratories and is suitable for routine bacterial culture and transformation experiments. It is a formulation of Luria-Bertani (LB) agar developed by Miller, which includes a mixture of peptides, amino acids, and vitamins to support the growth of a wide range of bacteria.

Oxoid Listeria Selective Enrichment Supplement is used for the enrichment of Listeria spp.

Listeria Selective Enrichment Supplement, Oxoid Composition

Listeria Selective Enrichment Medium Preparation:
Reconstitute one vial as directed, and add the contents to 500ml of  Listeria Enrichment Broth Base (CM0862). Sterilize by autoclaving at 121°C for 15 minutes. Cool to 50°C and mix well before distributing into sterile containers in volumes as required.

Listeria Selective Enrichment Supplement SR0141 as supplied should be stored at 2-8°C. When stored as directed the reagents are stable until the expiry date printed on the label.

Precautions
Listeria Selective Enrichment Supplement contains cycloheximide and is toxic if swallowed, inhaled or by skin contact. As a precaution when handling wear gloves and eye/face protection.
Acriflavine is activated by light and may form compounds inhibitory for Listeria.

Oxoid Yersinia Selective Supplement is used for the isolation of Yersinia enterocolitica.

Yersinia Selective Supplement, Oxoid Composition

Yersinia Selective Agar, Oxoid Preparation:
Suspend 29g in 500ml of distilled water and bring gently to the boil to dissolve completely. Sterilise by autoclaving at 121°C for 15 minutes. Allow to cool to approximately 50°C and aseptically add the contents of one vial of Yersinia Selective Supplement SR0109 reconstituted as directed in the instructions for use that accompany the product. Mix gently and pour into sterile Petri dishes.

Store the prepared medium at 2-8°C for not more than 24 hours.

Precautions
Some strains of Yersinia enterocolitica may grow poorly or not at all. Other species of Yersinia may grow along with some enteric organisms. It is therefore essential that full identification tests are carried out on suspect colonies.
