Membrane Endo Agar LES, Oxoid Preparation:
Suspend 45 grams in 1 litre of distilled water. Add 12ml of a 10% w/v solution of Basic Fuchsin. Heat gently with frequent agitation until the medium boils. DO NOT AUTOCLAVE. Cool to 45°C and dispense into 50-60 mm dishes in 4 ml volumes. For larger dishes use sufficient medium to give an equivalent depth (approx. 1.5 mm).
Plates should be protected from light and may be stored for up to two weeks in the refrigerator.

Store the dehydrated medium below 25°C and use before the expiry date on the label.
Store the prepared medium in the dark and at 2-8°C.

Aseptically add 10ml of distilled water to one vial and invert gently to dissolve. Add the vial contents to 2.25 litres of Listeria Enrichment Broth Base. Sterilize by autoclaving at 121°C for 15 minutes. Cool to 50°C, mix well and distribute into sterile containers in volumes as required .

Agar Preparation:
To Prepare the Agar Base:
Suspend 24.2g of the agar base, in 500ml of distilled water. Add 5ml of glycerol. Bring to the boil to dissolve completely, sterilise by autoclaving at 121°C for 15 minutes. Allow the medium to cool to 50°C.
To Prepare Pseudomonas CN Agar:
To 500ml of agar base cooled to 50°C add the contents of 1 vial of Pseudomonas CN Supplement (SR0102) rehydrated as directed. Mix well and pour into sterile Petri dishes.
To Prepare Pseudomonas CFC Agar:
To 500ml of agar base cooled to 50°C add the contents of 1 vial of Pseudomonas CFC Supplement (SR0103) rehydrated as directed. Mix well and pour into sterile Petri dishes.

Precautions
Fresh media should be prepared as required. Molten agar should not be kept longer than 4 hours. Medium should not be stored and remelted. If swarming colonies of Proteus species are a problem in food samples then the incubation temperature can be lowered to 20°C for a period of 3-5 days. Chilled foods may carry a wide range of pseudomonads and the colonies on CFC Medium, incubated at lower temperatures, may be Pseudomonas fluorescens or Pseudomonas putida as well as Pseudomonas aeruginosa. Aeromonas species will also appear as pink/brown colonies, particularly from fish products.

TBE buffer (Tris-Borate-EDTA, Biobasic) is commonly used in all DNA electrophoresis applications (both for acrylamide and for agarose gels). In general, TBE buffer offers better resolution of 0.1 to 3 kb fragments; whereas, TAE (Tris-Acetate-EDTA) buffer provides better resolution of fragments greater than 4 kb. Furthermore, TBE is better suited for high-voltage (>150V) electrophoresis because of its higher buffering capacity and lower conductivity than TAE. SeaKem® LE Agarose was the first and is still the world’s most trusted agarose for nucleic acid electrophoresis. Ideal for routine analysis of nucleic acids by gel electrophoresis and blotting, each SeaKem® LE Gel sharply resolves DNA and provides consistent resolution from lot-to-lot. This molecular biology grade agarose has no detectable DNase or RNase activity and forms strong gels with low background upon ethidium bromide, SYBR® Green, or GelStarTM Staining. Due to its low EEO, DNA will have a high electrophoretic mobility. SeaKem® LE Agarose is also recommended for protein gel electrophoresis in applications such as Ouchterlony and radial immunodiffusion (RID).
