Bordetella Selective Supplement, Oxoid Composition

Bordetella Selective Medium, Oxoid Preparation:
Aseptically add 2ml of sterile distilled water to one vial and mix gently to dissolve the contents completely. Aseptically add the contents to 500ml of sterile molten Charcoal Agar CM0119, cooled to approximately 50°C, together with 10% defibrinated horse blood SR0050 . Mix well and pour into sterile Petri dishes.
The vial contents may also be added to half strength Charcoal Agar CM0119 plus 10% defibrinated horse blood SR0050, for use as a transport medium for Bordetella pertussis.

Enrich and isolate halophilic organisms, especially staphylococci, using Thermo Scientific™ Oxoid™ Salt Meat Broth Tablets. Salt Meat Broth Tablets are specially designed for the isolation of staphylococci from grossly contaminated specimens such as feces, particularly during the investigation of staphylococcal food poisoning. The medium is highly sensitive as it can detect even low numbers of staphylococci from samples having large proportions of heterogeneous microbial flora.^1,2

Salt Meat Broth, Oxoid Preparation:
Add 2 tablets to 10ml of distilled water in an appropriately sized test tube and soak for 5 minutes. Sterilise by autoclaving at 121°C for 15 minutes.

Muller-Kauffmann Tetrathionate-Novobiocin Broth (MkTTn), Oxoid Preparation:
Suspend 89.5g of MKTTn in 1 litre of distilled water, mix well and bring to the boil. Cool to below 45°C. Immediately before use add 20ml of iodine-iodide solution prepared by dissolving 25g of potassium iodide in 10ml of water, adding 20g of iodine and then diluting to 100ml with sterile water. Also add the contents of four vials of Oxoid Novobiocin Selective Supplement SR0181 reconstituted as directed. Mix well and aseptically dispense into sterile containers.

Store the prepared media at room temperature. The prepared medium in its final form containing iodine/iodide solution + Novobiocin Selective Supplement SR0181 should be used immediately.

Oxoid Rogosa Agar is used for the selective isolation and enumeration of lactobacilli.

Rogosa Agar, Oxoid Preparation:
Suspend 82g in 1 litre of distilled water and bring to the boil to dissolve completely. Add 1.32ml glacial acetic acid and mix thoroughly. Heat to 90-100°C for 2-3 minutes with frequent agitation. Distribute into sterile tubes, Petri dishes or bottles.
DO NOT AUTOCLAVE.

Precautions
Lactobacillus carnis does not grow on this medium.
All colonies must be tested by Gram stain and catalase test before carrying out further identification tests.
