Campylobacter Agar (Karmali), Oxoid Preparation:
Add 21.5g of Campylobacter Agar Base (Karmali) to 500ml of distilled water and bring to the boil to dissolve. Sterilise by autoclaving at 121°C for 15 minutes. Cool to 50°C. Aseptically add 1 vial of Campylobacter Selective Supplement (Karmali) SR0167 or Modified Karmali Selective Supplement SR0205, reconstituted as directed.. Mix well and pour into sterile Petri dishes.

Campylobacter Agar Base (Karmali) should be stored tightly capped in the original container in a cool, dry place away from bright light. When stored at 10-30°C the medium will remain stable until the expiry date printed on the label.

Lysine Medium, Oxoid Preparation:
Suspend 6.6g in 100ml distilled water containing 1.0ml Potassium lactate SR0037. Bring to the boil to dissolve completely. Agitate frequently to prevent superheating. Cool to 50°C and add 0.1ml of lactic acid 10% SR0021 to adjust to pH 4.8 ± 0.2. Dispense into Petri dishes and remove surface moisture by drying at 37°C.

Description
Myosin phosphatase target subunit 1 (MYPT1), also called Myosin-binding subunit of Myosin phosphatase, is one of the subunits and an integral component of the Myosin phosphatase. Myosin phosphatase regulates the interaction of Actin and Myosin downstream of the guanosine triphosphatase Rho, which inhibits Myosin phosphatase through the action of Rho-kinase. It is also involved in smooth muscle contraction. Inhibition of Myosin light chain phosphatase results in Ca2+ sensitization of smooth muscle contraction. This inhibition is modulated through phosphorylation of MYPT1 by a ZIP-like kinase, which associates with MYPTI and phosphorylates the inhibitory site in smooth muscle. The phosphorylation of MYPT1 by protein kinase C results in altered dephosphoryation of Myosin by attenuating the binding of protein phosphatase 1 catalytic subunit (PP1c) and the phosphorylated 20 kDa Myosin light chain to MYPT1. PP1c interacts with at least four binding sites on the amino-terminus of MYPT1. MYPT1 is localized on stress fibers, and is distributed close to the cell membrane and at cell-cell contacts to regulate Myosin phosphatase activity.

Trichomonas Medium, Oxoid Preparation:
Suspend 37.5g in 1 litre of distilled water and bring to the boil to dissolve. Sterilise by autoclaving at 121°C for 15 minutes. Cool to approximately 50°C. Inactivate 80ml of horse serum, adjust to pH 6.0 and add it to the medium. For diagnostic work, bacterial growth may be suppressed by the addition of 1000 units of penicillin and 500µg of streptomycin per ml of medium or 100µg of chloramphenicol per ml of medium (1 vial of SR0078 per 500ml medium).
Prior to incorporation in the medium, Horse Serum SR0035 is inactivated by maintaining at a temperature of 56°C for 30 minutes, and then acidified with 1N hydrochloric acid to pH 6.0.

SeaKem® LE Agarose was the first and is still the world’s most trusted agarose for nucleic acid electrophoresis. Ideal for routine analysis of nucleic acids by gel electrophoresis and blotting, each SeaKem® LE Gel sharply resolves DNA and provides consistent resolution from lot-to-lot. This molecular biology grade agarose has no detectable DNase or RNase activity and forms strong gels with low background upon ethidium bromide, SYBR® Green, or GelStarTM Staining. Due to its low EEO, DNA will have a high electrophoretic mobility. SeaKem® LE Agarose is also recommended for protein gel electrophoresis in applications such as Ouchterlony and radial immunodiffusion (RID).
