Suspend 66.73g in 1 litre of distilled water. Boil to dissolve the medium completely. Dispense into tubes, bottles or flasks and sterilize by autoclaving at 121°C for 15 minutes. Selectivity can be altered by pH adjustment.

Store the prepared plates at 2–8°C.
Precautions
Sterilization of this medium in some autoclaves, at 121°C for 15 minutes, may cause the medium in some cases to fall outside the specified pH limit of 5.7 ± 0.1. Adjustment of the medium in these rare cases is recommended using acetic acid or sodium hydroxide.

Precautions
Brucella cultures are highly infective and must be handled under properly protected conditions. Incubate in 5-10% carbon dioxide atmosphere for 24-48 hours.
Campylobacter species are best grown at 42°C (except Campylobacter fetus subsp. fetus) in a micro-aerophilic atmosphere (Oxoid Campylobacter Gas Generating Kit BR0056 or BR0060 or CampyGen CN0025/CN0035).
Staph./Strep. supplemented plates should be incubated aerobically at 35°C for 18 hours. Incubation in carbon dioxide- enriched air will cause inhibition of staphylococcal growth.
Streptococcus Selective Supplement SR0126 supplemented plates may be incubated aerobically or anaerobically at 35°C for 18 hours.
Prepared plates of both supplemented media should be used within 18 hours of preparation for maximum selectivity. Gardnerella supplemented plates should be incubated at 35°C for 48 hours in an atmosphere containing 7% carbon dioxide.
Carry out confirmatory tests on all colonies from horse blood medium and on beta-haemolytic colonies from human or rabbit blood medium.
Incubate plates of Clostridium E-Y Agar anaerobically at 35°C for 18 hours, look for lecithinase activity (pearly layer) and for proteolysis. Lecithinase activity is inhibited in the presence of specific anti-toxin.

Precautions
Regard all suspicious colonies as Staphylococcus aureus regardless of negative reactions in the medium and carry out further tests.

Egg Yolk Emulsion, Oxoid Directions:
Suspend 21.5g of MYP Agar (CM0929) in 450ml of distilled water and bring gently to the boil to
dissolve. Sterilize by autoclaving at 121°C for 15 minutes. Cool to approximately 50°C and
aseptically add 50ml of Egg Yolk Emulsion (SR0047C) and the contents of one vial of Polymyxin B
Supplement (SR0099E) reconstituted as directed. Mix well and pour into sterile Petri dishes.
Alternatively, suspend 8g of Blood Agar Base (CM0055) in 200ml of distilled water and add 2g of
sodium chloride. Sterilize by autoclaving at 121°C for 15 minutes. Cool to approximately 50°C and
aseptically add 20ml Egg Yolk Emulsion (SR0047C). Mix well and pour into sterile Petri dishes.

Keep at temperatures between 2° and 8 °C. Use before expiry date on the label.
Precautions
On this medium Bacillus cereus is indistinguishable from Bacillus thuringiensis.
Identify Bacillus cereus by colony form, colour, egg yolk hydrolysis and confirm with cell and spore morphology.
Occasional strains of Bacillus cereus show weak or negative egg yolk reactions.

Buffer tablets pH 7.2 – for preparing buffer solution acc. to WEISE for staining of blood smears, are used to prepare buffer solutions according to WEISE and pH-buffered staining solutions. Phosphate buffer solutions are used in hematology, for the dilution of staining solutions and for the rinsing steps in the staining process. The buffer tablets acc. to Weise will produce a buffered solution (pH6.4/pH6.8/pH7.2) that stabilizes the staining result and gives reproducible staining results. Buffer solution acc. to Weise pH 6.4 results in bright-orange stained erythrocytes and differentiated stained cells with nuclei, whereby the erythrocytes are stained orange when using pH 6.8. Suspend 5.6g of ONE Broth-Salmonella Base in 225ml of distilled water. Sterilize by autoclaving at 121°C for 15 minutes. Cool to below 50°C and add the contents of 1 vial of ONE Broth-Salmonella Supplement (SR0242), resuspended as directed.
Storage conditions and Shelf life
The ONE Broth-Salmonella Selective Supplement should be stored in the dark at 2-8ºC. When stored as directed, the unopened products will remain stable until the expiry date printed on the packaging.
Once prepared, ONE Broth-Salmonella can be stored for up to 2 weeks when made from CM1091 and SR0242 according to the manufacturer’s instructions and stored at 2-8ºC, in the dark. A longer shelf life may be attainable, but should be validated under the relevant, local manufacturing and storage conditions.
