STAA Agar, Oxoid Preparation:
Suspend 18.5g of STAA Agar Base in 500ml distilled water and bring gently to the boil to dissolve completely. Add 7.5g of glycerol and sterilise by autoclaving at 121° C for 15 minutes. Cool to 50°C and aseptically add:
a) for STAA Medium the contents of 1 vial of STAA Selective Supplement SR0151 reconstituted as directed.
b) for STA Medium the contents of 1 vial of STA Selective Supplement SR0162 reconstituted as directed.
Mix well and distribute into sterile Petri dishes.

STAA Selective Medium Preparation:

Cool to 50°C and aseptically add the contents of 1 vial of STAA Selective Supplement SR0151 reconstituted as directed.

Precautions
STAA Selective Supplement SR0151 contains cycloheximide and thallous acatate and is toxic if swallowed, inhaled or if in contact with skin.

Precautions
Enterococcus faecalis may grow on this medium as tiny colonies with slight mannitol fermentation.
The high salt content in Staphylococcus Medium No.110 may interfere with the coagulase reaction. Always sub-culture to a non-inhibitory medium before testing.

Stuart Transport Medium, Oxoid Preparation:
Suspend 16g in 1 litre of distilled water. Bring to the boil to dissolve completely and dispense into screw-capped 7ml bottles. Fill each bottle to the brim, tighten the cap and sterilise by autoclaving at 121°C for 15 minutes. When sufficiently cool to handle, mix by inversion.

Precautions
A small amount of blue colour at the top of the bottle indicates oxidation. If this colour extends down into the medium it should be discarded.
Avoid prolonged heating in open flasks, during the preparation of the medium, because thioglycollate is volatile.
Sodium glycerophosphate may be metabolised by some organisms and thus promote their growth.

Selectively isolate pathogenic Vibrio species with Thermo Scientific™ Oxoid™ TCBS (Thiosulfate Citrate Bile Salts Sucrose) Cholera Medium (Dehydrated). The medium has shown to be the most satisfactory and reproducible formula regardless of the complex composition. Unlike Lauryl Sulphate Tellurite Agar it does not require further additives or aseptic additions of blood. It also possesses superior growth characteristics for Vibrio species, compared with tellurite media. While inhibiting non-vibrios, it promotes rapid growth of pathogenic vibrios after overnight incubation at 35°C.

Precautions
The identification of the various Vibrio species on TCBS Medium is presumptive and further tests are required for confirmation.

Colonies taken from TCBS Medium are `sticky’ and react poorly in slide agglutination tests. Subculture to nutrient agar is required before slide agglutination tests can be carried out.
Some strains of Vibrio vulnificus produce better recovery at 30°C.

Tetrathionate Broth is recommended for the selective enrichment method of isolating Salmonella typhi and other salmonellae from faeces, sewage, etc.
Organisms which reduce tetrathionate, such as salmonellae, flourish in the medium whilst many faecal organisms are inhibited. Members of the Proteus group reduce tetrathionate and may consequently impair the value of this medium for the isolation of salmonellae; this disadvantage of the medium is largely overcome by the addition of 40µg of novobiocin to each millilitre of the incomplete medium before the addition of iodine.

Suspend 21.5g of MYP Agar (CM0929) in 450ml of distilled water and bring gently to the boil to
dissolve. Sterilize by autoclaving at 121°C for 15 minutes. Cool to approximately 50°C and
aseptically add 50ml of Egg Yolk Emulsion (SR0047C) and the contents of one vial of Polymyxin B
Supplement (SR0099E) reconstituted as directed. Mix well and pour into sterile Petri dishes.
Alternatively, suspend 8g of Blood Agar Base (CM0055) in 200ml of distilled water and add 2g of
sodium chloride. Sterilize by autoclaving at 121°C for 15 minutes. Cool to approximately 50°C and
aseptically add 20ml Egg Yolk Emulsion (SR0047C). Mix well and pour into sterile Petri dishes.

Precautions
On this medium Bacillus cereus is indistinguishable from Bacillus thuringiensis.
Identify Bacillus cereus by colony form, colour, egg yolk hydrolysis and confirm with cell and spore morphology.
Occasional strains of Bacillus cereus show weak or negative egg yolk reactions.

Precautions
This medium lacks agar and reducing indicator, therefore it is essential that the medium is freshly prepared and used within four hours of preparation.
Thioglycollate media should not be reheated more than once because toxic oxygen radicles are formed on reheating.

If the upper portion of the medium is red because of oxidation, anaerobic conditions can be restored by reheating for 10 minutes in boiling water or steam. Do not reheat more than once.
Following reheating, if more than one-third of the medium is oxidised then it should be discarded.
Some glucose-fermenting organisms which are able to reduce the pH of the medium to a critical level may not survive in this medium. Early subculture is necessary to isolate these organisms.

Thioglycollate Medium, Oxoid Preparation:
Suspend Thioglycollate medium (29.75 grams/ 1itre) in de-ionised water. Heat to dissolve. Cool and dispense 100ml into final containers, 100ml DIN bottles. Sterilise at 121°C for 15 minutes. When cooled label each bottle and pack in units of 10 into labelled boxes.
Storage conditions and Shelf life
Store away from light between 2-25ºC. Product shelf life from date of production is 1 year.

Oxoid Thioglycollate Medium, Brewer is an anaerobic medium especially useful for sterility control of solutions containing mercury-based preservatives.

Thioglycollate Medium, Brewer, Oxoid Preparation:
Suspend 20g in 1 litre of distilled water. Bring to the boil, mix well and allow to stand until completely dissolved. Distribute into final containers and sterilise by autoclaving at 121°C for 15 minutes.
Media containing small quantities of agar are liable to separate if cooled rapidly. Tubes of reconstituted and autoclaved medium should be allowed to cool slowly on a wooden surface in a draught-free atmosphere.
Storage conditions and Shelf life
Store the dehydrated medium at 10-30°C and use before the expiry date on the label.
The prepared medium should be stored away from light at room temperature.
Precautions
Check the upper portion of the prepared medium before inoculation. If more than one-third of the fluid is oxidised i.e. green coloured, discard the bottle.
If one-third or less fluid is oxidised, then heat in boiling water with the cap loosened to drive off oxygen. Cool to room temperature before inoculation. This reheating process can only be carried out once because of the formation of toxic radicles in the medium.
Organisms which ferment glucose and lower the pH to critical levels may not survive in this medium after growth has taken place.
