Instructions for Use:
Dissolve 4.54gms of dehydrated medium in 600ml of distilled or deionized water at room temperature (15-30*C). Rinse media vial with small quantity of distilled water to remove traces of power. Add the desired heat stable supplements prior to autoclaving. Continue stirring until the powder has dissolved. Some times media does not dissolve completely unless the pH is reduced. For these, lower the pH to about 3.0 to facilitate dissolution of media. The pH of medium is adjusted by using 1N HCL/ 1N NaOH/ 1NKOH. Make up the final volume to 1000ml with distilled water. Mix Gently, heat and rotate between intervals until the solution becomes clear. Do not boil, reheat and allow to cool below 50*C during dispensing. Dispense the medium into suitable containers, plug or cap, then autoclave at 15pso (121*C) for 15 minutes, using a slow exhaust cycle. Higher temperatures and/or longer times are not recommended. Cool the autoclaved culture vessels containing medium to 45-50*C and aseptically add desired sterile heat-labile substrate.
Note: Media should be prepared according to formula mentioned on the label, however, it is recommended to use an entire container at once. Heat-liable substrates should be added, after autoclaving.

Mycoplasma Agar, Oxoid Preparation:
Add 35.5g to 1 litre of distilled water. Boil to dissolve the agar and distribute in 80ml volumes. Sterilise by autoclaving at 121°C for 15 minutes. Cool to approximately 50°C and add one vial of Mycoplasma Supplement SR0059 reconstituted as directed.

Store the prepared medium at 2-8°C.
Precautions
Thallous acetate is toxic, observe the precautions in the HAZARDS section.
Do not use thallous acetate media to isolate Ureaplasma urealyticum.
Mycoplasmas may be suspected if (1) typical morphology (2) no growth in media without serum (3) colonies are embedded below the agar surface.

Mycoplasma Medium Preparation:
Prepare the sterile supplement by aseptically adding 20ml sterile distilled water to the vial and mix gently. Add 1ml of Oxoid Mycoplasma Base without supplements to each of ten small bottles. Sterilise by autoclaving at 121°C for 15 minutes. Allow to set. Aseptically add 2ml of the reconstituted Supplement-P to each bottle containing agar.

Selectively enumerate Bacillus cereus in food samples with Thermo Scientific™ Oxoid™ MYP Agar (Dehydrated). The medium relies upon mannitol fermentation and phospholipase C production to differentiate Bacillus cereus from other members of the species.
Mannitol-negative Bacillus cereus form pink colored colonies due to the presence of Phenol Red Indicator. Egg yolk emulsion helps in the differentiation of phospholipase C producing colonies, which are surrounded by a zone of the white precipitate. Polymixin B Sulphate acts as the inhibitor to restrict the growth of Gram-negative bacteria.

MYP Agar, Oxoid Preparation:
Suspend 21.5g in 450ml distilled water and bring gently to the boil to dissolve. Sterilise by autoclaving at 121°C for 15 minutes.
Cool to approximately 49°C and aseptically add 50ml Egg Yolk Emulsion SR0047 and 1 vial of Polymyxin B Supplement SR0099, reconstituted as directed. Mix well and pour into sterile petri dishes.

Selective Medium for Non-Sporing Anaerobes Preparation:
Suspend 21.5g of Wilkins-Chalgren Anaerobe Agar (CM0619) in 475ml of distilled water containing 0.5ml `Tween 80’. Bring to the boil to dissolve completely and sterilise by autoclaving at 121°C for 15 minutes. Cool to 50-55°C and aseptically add the contents of 1 vial of N-S Anaerobe Supplement (SR0107) rehydrated as directed, together with 25ml of defibrinated blood (SR0050/SR0051).
