Muller-Kauffmann Tetrathionate-Novobiocin Broth (MkTTn), Oxoid Preparation:
Suspend 89.5g of MKTTn in 1 litre of distilled water, mix well and bring to the boil. Cool to below 45°C. Immediately before use add 20ml of iodine-iodide solution prepared by dissolving 25g of potassium iodide in 10ml of water, adding 20g of iodine and then diluting to 100ml with sterile water. Also add the contents of four vials of Oxoid Novobiocin Selective Supplement SR0181 reconstituted as directed. Mix well and aseptically dispense into sterile containers.

Store the dehydrated medium at 10-30°C and use before the expiry date on the label.
Store the prepared media at room temperature. The prepared medium in its final form containing iodine/iodide solution + Novobiocin Selective Supplement SR0181 should be used immediately.

Brilliance E. coli/coliform Selective Agar, Oxoid Preparation:
Suspend 28.1g of Brilliance E. coli/coliform Selective Agar in 1 litre of distilled water. Bring the medium gently to the boil, to dissolve completely. Either pour the medium into sterile Petri dishes or keep molten at 45ºC for pour plate technique.

Suspend 20.5g in 500ml of distilled water. Mix well and bring to the boil to dissolve completely. Sterilize by autoclaving at 121°C for 15 minutes. Cool the medium to 50°C and aseptically add 1 vial of Brilliance Bacillus cereus Selective Supplement. Mix well and pour into sterile Petri dishes.

Precautions
X-glucopyranoside negative B. cereus strains may present white colonies on this medium due to poor or no utilisation of the chromogen.

Suspend 14.63g of Sodium Chloride Peptone Broth (Buffered) in 1 litre of water (purified as required). If required, add 1-10ml Tween® 20 or 80 per litre of medium. Mix well and distribute into final containers. Sterilize by autoclaving at 121°C for 15 minutes.

Precautions
Ensure that the glass tubes used for the preparation of the medium do not auto fluoresce and take the relevant safety precautions when using a UV lamp.
Some pathogenic strains, typically Escherichia coli O157:H7 are usually glucuronidase negative and therefore will not give a positive result in this test.
Other enteric organisms e.g. some Shigella species are also glucuronidase positive.
