Violet Red Bile Lactose Agar (VRBL) (ISO) (Dehydrated), Oxoid Preparation:
Suspend 38.5g of VRBL Agar (ISO) dehydrated medium in 1 litre of distilled water. Bring to the boil while mixing. Continue to boil for up to 2 minutes or for the minimum time necessary to dissolve completely. DO NOT AUTOCLAVE. Cool to 44-47°C and use within 4 hours. Mix well before pouring.

Storage conditions and Shelf life
VRBL Agar (ISO) should be stored in the tightly capped original container at 10-30°C. When stored as directed, the un-opened product will remain stable until the expiry date printed on the packaging.

Locally prepared VRBL Agar (ISO) plates can be stored for up to 2 weeks when made from CM0968 according to the manufacturer’s instructions and stored at 2-8°C, out of direct sunlight. A longer shelf life may be attainable, but should be validated under the relevant, local manufacturing and storage conditions.

Precautions
VRBL Agar (ISO) should be prepared, cooled to 44-47°C and used within 4 hours.

VRBL Agar (ISO) is for laboratory use only. Do not use the dehydrated medium beyond the stated expiry date or if the product shows any sign of deterioration.

Arcobacter Broth Base, Oxoid Preparation:

Dissolve 12g of Arcobacter Broth in 500ml of distilled water. Sterilise at 121°C for 15 minutes and allow to cool to below 50°C

For use with CAT supplement (SR0174)

Add one vial per 500ml medium of CAT Selective Supplement reconstituted as directed. Dispense into sterile containers.

For use with CCDA supplement (SR0155)

Add one vial per 500ml medium of CCDA Selective Supplement reconstituted as directed. Dispense into sterile containers.

Inoculated plates should be incubated aerobically, at 30°C, for 24 hours.

Baird-Parker Agar Base (RPF), Oxoid Preparation:
Suspend 6.3g of Baird-Parker Agar Base (RPF) in 90ml of distilled water. Bring to the boil to dissolve completely. Sterilise by autoclaving at 121ºC for 15 minutes. Cool to 48ºC and add 1 vial of SR0122A, reconstituted as directed. Mix well and pour plates.

Storage conditions and Shelf life
Store the dehydrated medium at 10-30°C and RPF Supplement below 0°C. Use before the expiry date on the label.
Prepared plates of medium are best used freshly prepared.

Precautions
Colonies of some contaminating organisms growing in close proximity to the coagulase positive colonies may partially digest the coagulase halo reaction.

Some pathogenic strains, typically Escherichia coli O157:H7 are usually glucuronidase negative and therefore will not give purple colonies on this medium.

TBX Medium 500g

Detect and enumerate Escherichia coli in food and animal feed samples with Thermo Scientific™ Oxoid™ Typtone Bile X-Glucuronide (TBX) Medium (Dehydrated). The medium meets the formulation and performance criterion laid down in ISO 16649 and ISO 11133:2014.

TBX Medium builds on the speed, reliability and recovery from frozen samples and detection of poor lactose fermenters offered by the original Tryptone Bile Agar, with the addition of a chromogenic agent, X-glucuronide, which detects glucuronidase activity, an enzyme shown to be highly specific for Escherichia coli.

MYP Agar 500g

Selectively enumerate Bacillus cereus in food samples with Thermo Scientific™ Oxoid™ MYP Agar (Dehydrated). The medium relies upon mannitol fermentation and phospholipase C production to differentiate Bacillus cereus from other members of the species.

Mannitol-negative Bacillus cereus form pink colored colonies due to the presence of Phenol Red Indicator. Egg yolk emulsion helps in the differentiation of phospholipase C producing colonies, which are surrounded by a zone of the white precipitate. Polymixin B Sulphate acts as the inhibitor to restrict the growth of Gram-negative bacteria.

Mix well and pour into sterile Petri dishes.

MSRV Medium, Oxoid Preparation:
Suspend 15.8g of MSRV Medium Base in 500ml of distilled water. Bring to the boil with frequent agitation.
DO NOT AUTOCLAVE. Cool to 50°C and aseptically add the contents of 1 vial of MSRV Selective Supplement SR0161E reconstituted with 2ml of sterile distilled water. Mix well and pour into sterile Petri dishes. Air dry at room temperature for at least one hour. (Plates may be air-dried overnight prior to storage at 2-8°C.)

Enumerate heterotrophic bacteria from drinking water samples with Thermo Scientific™ Oxoid™ R2A Agar (Dehydrated). This nutritionally reduced medium was specially designed to enhance the recovery of stressed and chlorine damaged bacteria from treated water samples. Stressed or injured organisms during water treatment are unable to grow on high nutrient media since faster-growing organisms outgrow the former. Therefore the use of a low nutrient medium like R2A Agar incubated for longer incubation periods allows these stressed organisms to grow well.

Fraser Broth, Oxoid Preparation:
With Fraser Supplement:
Suspend 28.7g in 500ml of distilled water and mix well to dissolve completely. Sterilise by autoclaving at 121°C for 15 minutes. Cool to below 50°C, and aseptically add the contents of one vial of Fraser Selective Supplement (SR0156) reconstituted as directed in the product insert. Mix well and distribute into sterile containers.

With Half Fraser Supplement:
Suspend 12.9g of Fraser Broth in 225ml of distilled water., and mix well to dissolve completely Sterilise by autoclaving at 121°C for 15 minutes. Cool to below 50°C, and aseptically add the contents of one vial of Half Fraser Selective Supplement SR0166E reconstituted as directed in the product insert. Mix well and distribute into sterile containers. Alternatively, to make 2.25 litres, use 129.0g of CM0895 and 1 vial of SR0166G.

Bile Aesculin Agar 500g

Isolate and presumptively identify enterococci / Group D streptococci with Thermo Scientific™ Oxoid™ Bile Aesculin Agar (Dehydrated). Bile Aesculin Agar is designed to differentiate between Group D streptococci and non-Group D streptococci.

Group D streptococci hydrolyse aesculin to form aesculetin and dextrose. Aesculin hydrolysis is indicative of a positive result. Bile salts in the medium inhibit Gram-positive bacteria other than enterococci/ Group D streptococci.

Precautions
RVS Broth should not be used if Salmonella Typhi is suspected.
