Suspend 45 g in 1 litre of distilled water. Bring to the boil to dissolve completely. Sterilise by autoclaving at 121°C for 15 minutes. Cool to 50°C and add 5% of inactivated Horse Serum (i.e. Horse Serum held at 56°C for 30 minutes). Mix well before pouring.

Trichomonas Medium, Oxoid Preparation:
Suspend 37.5g in 1 litre of distilled water and bring to the boil to dissolve. Sterilise by autoclaving at 121°C for 15 minutes. Cool to approximately 50°C. Inactivate 80ml of horse serum, adjust to pH 6.0 and add it to the medium. For diagnostic work, bacterial growth may be suppressed by the addition of 1000 units of penicillin and 500µg of streptomycin per ml of medium or 100µg of chloramphenicol per ml of medium (1 vial of SR0078 per 500ml medium).
Prior to incorporation in the medium, Horse Serum SR0035 is inactivated by maintaining at a temperature of 56°C for 30 minutes, and then acidified with 1N hydrochloric acid to pH 6.0.

To suppress the growth of bacteria, you can acidify the medium to pH 3.5 by adding 1 ml of lactic acid 10% SR0021 to each sterilized medium at 50°C. Note that the medium must not be heated after the addition of acid as it will destroy the gelling properties and results in hydrolysis of agar.

Charcoal Agar, Oxoid Preparation:
Suspend 51g in 1 litre of distilled water. Bring to the boil to dissolve completely. Sterilise by autoclaving at 121°C for 15 minutes. Cool to 50°C, add 10% of defibrinated blood and mix gently. The medium is made selective for the isolation of Bordetella pertussis and Bordetella parapertussis by the addition of Bordetella Selective Supplement SR0082.
To one vial add 2ml of sterile distilled water and dissolve the contents completely. Add this solution to 500ml of sterile, molten Charcoal Agar, cooled to 50°C, together with 10% v/v defibrinated horse blood SR0050. Mix well before pouring into sterile Petri dishes.
For Haemophilus influenzae, omit the selective agents and convert to `chocolate’ agar.

Precautions
Stuart’s transport medium or similar formulation media should not be used for Bordetella-containing specimens.
Two pernasal swabs should be taken from each patient, one through each nostril.
Make sure the charcoal remains in suspension when dispensing the medium by gently swirling the flask.
Lysed horse blood is used in the transport medium but whole blood is used in the isolation medium.
Most naso-pharyngeal flora are inhibited by cephalexin but Pseudomonas aeruginosa and some fungi may grow through. Amphotericin B can be added (12mg/ml) as an antifungal agent to prevent the growth of filamentous fungi. However, this level of amphotericin B can be inhibitory to Bordetella pertussis and should not be used routinely.

Precautions
A small amount of blue colour at the top of the bottle indicates oxidation. If this colour extends down into the medium it should be discarded.
Avoid prolonged heating in open flasks, during the preparation of the medium, because thioglycollate is volatile.
Sodium glycerophosphate may be metabolised by some organisms and thus promote their growth.

Detect and enumerate coliform organisms in water, food and dairy products with Thermo Scientific™ Oxoid™ Violet Red Bile Agar (Dehydrated). This lactose containing selective medium has been used for the determination of the coli-aerogenes content of water, milk and other dairy products, dairy equipment, and food products. Due to the addition of selective inhibitory components, neutral red, crystal violet, and bile salts, the medium provides selectivity against non-target Gram-positive bacteria.

Violet Red Bile Agar (Dehydrated), Oxoid Preparation:
Suspend 38.5g in 1 litre of distilled water. Bring to the boil. Continue to boil for 2 minutes or for the minimum time necessary to dissolve completely and ensure that there are no remaining flecks of unmelted agar. No further sterilisation is necessary or desirable. Mix well before pouring.
Storage conditions and Shelf life
Store the dehydrated medium at 10-30°C and use before the expiry date on the label.
Store the prepared medium at 2-8°C and use as freshly as possible.
Precautions
This medium is not completely specific for Enterobacteriaceae, other organisms e.g. Aeromonas and Yersinia species may give similar reactions.
The selectivity of the medium diminishes after 24 hours incubation and organisms previously suppressed may exhibit growth.

Corn Meal Agar, Oxoid Preparation:
Suspend 17g in 1 litre of distilled water. Bring to the boil to dissolve completely. Sterilise by autoclaving at 121°C for 15 minutes.

Store the prepared medium at 2-8°C.
Precautions
Glucose supplemented Corn Meal Agar should not be used for chlamydospore production.
Corn Meal Agar with `Tween 80’ (or other wetting agents) will allow Candida stellatoides and Candida tropicalis to produce chlamydospores.
Some Candida strains lose their ability to produce chlamydospores after repeated subculturing.

Isolate and identify Salmonella and Shigella species from clinical or food samples with Thermo Scientific™ Oxoid™ SS Agar (Dehydrated). Differentiation of Salmonella and Shigella is achieved by incorporation of lactose in the medium. Organisms which ferment lactose produce acid which, in the presence of the neutral red indicator, results in the formation of red colonies. Salmonella, Shigella, and other non-lactose-fermenting organisms appear as transparent or straw coloured colonies on SS Agar.

SS Agar, Oxoid Preparation:
Suspend 63g in 1 litre of distilled water. Bring to the boil with frequent agitation and allow to simmer gently to dissolve the agar. DO NOT AUTOCLAVE. Cool to about 50°C, mix and pour into sterile Petri dishes.

Store the prepared medium at 2-8°C.
Precautions
This medium is highly selective and R-strains of shigellae will not grow on it. It is not recommended for the primary isolation of shigellae.

Czapek Dox Agar Modified, Oxoid Preparation:
Suspend 45.4g in 1 litre of distilled water. Bring to the boil to dissolve completely. Sterilise by autoclaving at 121°C for 15 minutes. Mix well before pouring. If required, adjust the reaction to pH 3.5 ± 0.2 by adding 10ml of Lactic Acid 10% (SR0021) per litre after sterilisation.
