Malt Extract Agar, Oxoid Preparation:
Suspend 50g in 1 litre of distilled water and boil to dissolve. Sterilise by autoclaving at 115°C for 10 minutes.

If it is desired to adjust the medium to pH 3.5, cool to 55°C and add approximately 2-3ml of 10% Lactic Acid SR0021 to 100ml Malt Extract Agar. Once acidified with lactic acid, the medium should not be re-heated.

Precautions
It is important not to carry over any nutrients into the citrate medium because this will result in false positive tests. Dilute the inoculum in saline before inoculating the citrate medium to avoid a carry-over of other carbon sources.

Tetrathionate Broth Base 500g, Oxoid

Oxoid Tetrathionate Broth Base is used for selective enrichment of Salmonella species.

Tetrathionate Broth Base, Oxoid Composition
Add 77g to 1 litre of distilled water and bring to the boil. Cool below 45°C and add 20ml of iodine solution. Mix well and tube in 10ml quantities. The prepared base will keep for several weeks at 4°C but should be used soon after the addition of the iodine solution.

Tetrathionate Broth Preparation:
Tetrathionate Broth is recommended for the selective enrichment method of isolating Salmonella typhi and other salmonellae from faeces, sewage, etc.

Organisms which reduce tetrathionate, such as salmonellae, flourish in the medium whilst many faecal organisms are inhibited. Members of the Proteus group reduce tetrathionate and may consequently impair the value of this medium for the isolation of salmonellae; this disadvantage of the medium is largely overcome by the addition of 40µg of novobiocin to each millilitre of the incomplete medium before the addition of iodine.

Thioglycollate Medium (USP) (Dehydrated), Oxoid Preparation:
Suspend 29.75 g in 1 litre of distilled water. Bring to the boil to dissolve completely. Distribute into final containers, sterilise by autoclaving at 121°C for 15 minutes. Mix well and cool to room temperature.

Store the prepared medium away from light at room temperature.

Precautions
If the upper portion of the medium is red because of oxidation, anaerobic conditions can be restored by reheating for 10 minutes in boiling water or steam. Do not reheat more than once.
Following reheating, if more than one-third of the medium is oxidised then it should be discarded.
Some glucose-fermenting organisms which are able to reduce the pH of the medium to a critical level may not survive in this medium. Early subculture is necessary to isolate these organisms.

THIS MEDIUM IS VERY HYGROSCOPIC AND MUST BE PROTECTED FROM MOISTURE.
The quantities given for the formula as classically described made 1110ml of medium. They have been published this way in the Oxoid literature to coincide with the scientific literature.

The directions for reconstituting Oxoid Rappaport-Vassiliadis (RV) Enrichment Broth follow usual Oxoid practice and specify the weight needed for 1 litre of medium.

Rappaport-Vassiliadis Enrichment Broth (Dehydrated), Oxoid Preparation:
Add 30g (the equivalent weight of dehydrated medium per litre) to 1 litre of distilled water. Heat gently until dissolved completely. Dispense 10ml volumes into screw-capped bottles or tubes and sterilise by autoclaving at 115°C for 15 minutes.

Precautions
RV Broth should not be used if Salmonella typhi is suspected.

OGYE Selective Medium Preparation:

Allow the medium to cool to 50°C and aseptically add the contents of one vial of Oxytetracycline GYE Selective Supplement (SR0073) reconstituted as directed. Tryptone Soya Agar 500g, Oxoid

First, you add 40g of tryptone soya agar to 1 litre of distilled water (purified as required). You can boil to dissolve the powder completely. After dissolved, you can sterilise the medium by autoclaving at 121°C for 15 minutes.
