Edwards Medium, Modified, Oxoid Preparation:
Suspend 41g in 1 litre of distilled water. Bring to the boil to dissolve completely. Sterilise by autoclaving at 115°C for 20 minutes. Cool to 50°C, add 5-7% of sterile bovine or sheep blood, mix well and pour plates.

Bile Aesculin Agar 500g

Isolate and presumptively identify enterococci / Group D streptococci with Thermo Scientific™ Oxoid™ Bile Aesculin Agar (Dehydrated). Bile Aesculin Agar is designed to differentiate between Group D streptococci and non-Group D streptococci.

Group D streptococci hydrolyse aesculin to form aesculetin and dextrose. Aesculin hydrolysis is indicative of a positive result. Bile salts in the medium inhibit Gram-positive bacteria other than enterococci/ Group D streptococci.

First, you add 40g of tryptone soya agar to 1 litre of distilled water (purified as required). You can boil to dissolve the powder completely. After dissolved, you can sterilise the medium by autoclaving at 121°C for 15 minutes.

Yeast and Mold Agar, Oxoid Preparation:
Suspend 41.0g in 1 litre of distilled water and bring to the boil to dissolve. Sterilise by autoclaving at 121°C for 15 minutes. The medium may be rendered selective after sterilisation by acidifying to pH 4.0 with 12-15ml of Lactic Acid SR0021 after cooling to 50°C. Do not reheat after making this addition. Mix well and pour into sterile Petri dishes.

Trichomonas Medium, Oxoid Preparation:
Suspend 37.5g in 1 litre of distilled water and bring to the boil to dissolve. Sterilise by autoclaving at 121°C for 15 minutes. Cool to approximately 50°C. Inactivate 80ml of horse serum, adjust to pH 6.0 and add it to the medium. For diagnostic work, bacterial growth may be suppressed by the addition of 1000 units of penicillin and 500µg of streptomycin per ml of medium or 100µg of chloramphenicol per ml of medium (1 vial of SR0078 per 500ml medium).

Prior to incorporation in the medium, Horse Serum SR0035 is inactivated by maintaining at a temperature of 56°C for 30 minutes, and then acidified with 1N hydrochloric acid to pH 6.0.

Pre-supplemented Dichloran-Glycerol 18% (DG18) (ISO) Agar Base, Oxoid Preparation:
Suspend 15.8g per 500ml (31.6g/l) of distilled water and heat to dissolve completely. Before autoclaving add 220g of Glycerol per 1 litre of medium. Heat the complete solution gently to aid its dissolution of the glycerol. This has been found to be a key procedure to follow to allow proper dissolution of the glycerol. Sterilize by autoclaving at 121°C for 15 minutes. Cool to 50°C, mix well and pour into sterile Petri dishes.

Where bacterial overgrowth occurs from certain foods (raw meats) chlortetracycline hydrochloride at 50mg/L (dissolved in water and filter sterilised) is recommended.

Precautions
The dichloran compound used in this medium is Botran® 2,6-Dichloro-4-Nitro-Analine (CAS: 99-30-9).

Pre-supplemented DG18 (ISO) Agar (CM1151) contains Chloramphenicol in the base powder and is therefore considered a toxic compound which should be handled in accordance with good industrial hygiene and safety practice. Always consult the MSDS before handling the powder.

The china blue in the medium serves as a pH indicator to differentiate lactose fermenters from non-lactose fermenters. The medium does not contain any inhibitory substances therefore all the organisms present in milk sample grow luxuriantly on this medium.

Cary-Blair Transport Medium, Oxoid Preparation:
Suspend 13.3g in 1 litre of distilled water and bring gently to the boil to dissolve the agar. Distribute into small, screw-cap bottles and sterilise by immersing in free-steam for 15 minutes. Allow to cool and tighten the screw caps to prevent water loss.

The prepared medium should be stored away from light at 2-8°C or at room temperature (22-25°C) up to 19 months.

Precautions
The medium should not be incubated to check sterility, prior to use. This should be carried out on separate quality control samples.
The medium can maintain the viability of fastidious organisms for transport purposes but it should not be used as a storage or enrichment medium.
The results obtained from the medium are dependent on the quality of the specimen material. Commensal anaerobic organisms may overgrow in the medium and cause misleading results.

Aseptically add 25ml of Lysed Horse Blood (SR0048), and 1 vial of Preston Campylobacter Selective Supplement (SR0117 or SR0204) reconstituted as directed and one vial of Campylobacter Growth Supplement (SR0232). Mix well and pour into sterile Petri dishes.

Preston Campylobacter Selective Enrichment Broth, Oxoid Preparation:
Dissolve 12.5g of Nutrient Broth No.2 (CM0067) in 475ml of distilled water and sterilise by autoclaving at 121°C for 15 minutes. Cool to 50°C or below. Aseptically add 25ml of Lysed Horse Blood (SR0048), 1 vial of Preston Campylobacter Selective Supplement (SR0117 or SR0204) and 1 vial of Campylobacter Growth Supplement (SR0232). Aseptically dispense 5ml volumes in sterile small screw-capped bottles. The Selective Enrichment Broth may be stored for up to 7 days at 2-8°C.

DNase agar 500g

Detect microbial deoxyribonuclease enzymes, particularly from staphylococci, with Thermo Scientific™ Oxoid™ DNASE Agar (Dehydrated). A DNase (deoxyribonuclease) is any enzyme that catalyzes the cleavage of phosphodiester linkages of DNA leading to its degradation. The organisms producing DNase enzymes in sufficient quantities to hydrolyze DNA will show clear zones around the colonies. The medium is mainly used in the identification of staphylococci but may also be used for the detection of DNase activity in other organisms.

Precautions
The DNase reaction for staphylococci is an indication of pathogenicity, it cannot be used as the sole criterion for identification.
Small zones of clearing may be caused by other enzymes or organic acid production.
Other organisms than staphylococci, Serratia and aeromonads can produce DNases.
Once the hydrochloric acid has been applied to the medium the plate must be read within a few minutes and further testing cannot be carried out by re-incubation.
The methyl green must be purified by extraction with chloroform.
Toluidine blue varies in performance according to source.
Merck Toluidine blue 1273 is satisfactory. Note that this dye cannot be used for Gram positive organisms.

Sabouraud Maltose Agar, Oxoid Preparation:
Suspend 65g in 1 litre of distilled water. Bring to the boil to dissolve completely. Sterilise by autoclaving at 121°C for 15 minutes. Mix well and pour in to sterile Petri dishes.

Storage conditions and Shelf life
Store the dehydrated medium at 10-30°C and use before the expiry date on the label.
Store the prepared medium at 2-8°C.

Precautions
Some of the pathogenic fungi may produce infective spores which are are easily dispersed into the laboratory. Such organisms should be examined only within a protective cabinet.
The combination of cycloheximide and chloramphenicol inhibits many pathogenic fungi. However, the mycelial phase of Histoplasma capsulatum, Paracoccidioides brasiliensis, Sporothrix schoenckii and Blastomyces dermatitidis is not inhibited by these antibiotics when incubated at 25-30°C .

Selectively enrich Salmonella spp. from food, animal feces, and environmental samples with Thermo Scientific™ Oxoid™ Modified Semi-solid Rappaport Vassiliadis (MRSV) Agar (ISO) (Dehydrated). The efficacy of the medium is based upon the ability of Salmonellae to migrate through the selective medium ahead of competing Gram-negative, motile organisms and, thus, produce opaque halos of growth.

MSRV (ISO) Agar, Oxoid Preparation:
Suspend 31.6g of MSRV (ISO) Agar base in 1 litre of distilled water. Bring to the boil with frequent agitation. DO NOT AUTOCLAVE. Cool to 50°C, and aseptically add the contents of 1 vial of Novobiocin Supplement (SR0181), reconstituted as directed. Mix well, and pour into sterile Petri dishes. This medium is semi-solid, therefore do not invert plates.

Storage conditions and Shelf life
MSRV (ISO) should be stored in the tightly capped original container at 10-30°C. Novobiocin Supplement should be stored in the dark at 2-8°C. When stored as directed, the un-opened products will remain stable until the expiry date printed on the packaging.

Precautions
MSRV (ISO) Agar is for laboratory use only. Do not use the dehydrated medium beyond the stated expiry date or if the product shows any sign of deterioration.
Basal medium is very hygroscopic.
