Lysine Iron Agar, Oxoid Preparation:
Suspend 34g in 1 litre of distilled water. Bring to the boil to dissolve completely. Dispense into tubes and sterilise by autoclaving at 121° C for 15 minutes. Cool the tubes in an inclined position to form slants with deep butts.

MYP Agar 500g

Selectively enumerate Bacillus cereus in food samples with Thermo Scientific™ Oxoid™ MYP Agar (Dehydrated). The medium relies upon mannitol fermentation and phospholipase C production to differentiate Bacillus cereus from other members of the species.

Mannitol-negative Bacillus cereus form pink colored colonies due to the presence of Phenol Red Indicator. Egg yolk emulsion helps in the differentiation of phospholipase C producing colonies, which are surrounded by a zone of the white precipitate. Polymixin B Sulphate acts as the inhibitor to restrict the growth of Gram-negative bacteria.

MYP Agar, Oxoid Preparation:
Suspend 21.5g in 450ml distilled water and bring gently to the boil to dissolve. Sterilise by autoclaving at 121°C for 15 minutes.
Cool to approximately 49°C and aseptically add 50ml Egg Yolk Emulsion SR0047 and 1 vial of Polymyxin B Supplement SR0099, reconstituted as directed. Mix well and pour into sterile petri dishes.

Thioglycollate Broth Usp Alternative

Whilst storage of the prepared medium is not recommended, autoclaved volumes of Thioglycollate Medium USP – Alternative, should be held at 20-30°C in the dark. Storage at lower temperatures increases oxygen absorption.

Precautions
This medium lacks agar and reducing indicator, therefore it is essential that the medium is freshly prepared and used within four hours of preparation.
Thioglycollate media should not be reheated more than once because toxic oxygen radicles are formed on reheating.

Desoxycholate Agar 500g

Precautions
Sterilization of this medium in some autoclaves, at 121°C for 15 minutes, may cause the medium in some cases to fall outside the specified pH limit of 5.7 ± 0.1. Adjustment of the medium in these rare cases is recommended using acetic acid or sodium hydroxide.

Buffer tablets pH 7.2 – for preparing buffer solution acc. to WEISE for staining of blood smears, are used to prepare buffer solutions according to WEISE and pH-buffered staining solutions. Phosphate buffer solutions are used in hematology, for the dilution of staining solutions and for the rinsing steps in the staining process. The buffer tablets acc. to Weise will produce a buffered solution (pH6.4/pH6.8/pH7.2) that stabilizes the staining result and gives reproducible staining results. Buffer solution acc. to Weise pH 6.4 results in bright-orange stained erythrocytes and differentiated stained cells with nuclei, whereby the erythrocytes are stained orange when using pH 6.8. Buffer solution acc. to Weise pH 7.2 produces grayish stained erythrocytes and intense red-violet stained cells with nuclei.
100 tablets are sufficient for 100 l buffer solution, each produced solution can be stable up to 4 weeks. Buffer tablets acc. to Weise are IVD products and CE registered.

Precautions

Precautions
It is essential that Kligler Iron Agar is examined and reported at 18-24 hours. Early or late readings will give false results.
KIA will grow both oxidative and fermentative organisms. Confusion will result if care is not taken to distinguish between the two groups.
Always use a straight wire to inoculate the butt, to avoid splitting the agar with a loop.
Pure cultures are essential to avoid misinterpretation.
Do not use screw-capped tubes or bottles for KIA medium. It is essential that air is freely available to growth on the slant.

Agar Preparation:
To Prepare the Agar Base:
Suspend 24.2g of the agar base, in 500ml of distilled water. Add 5ml of glycerol. Bring to the boil to dissolve completely, sterilise by autoclaving at 121°C for 15 minutes. Allow the medium to cool to 50°C.

To Prepare Pseudomonas CN Agar:
To 500ml of agar base cooled to 50°C add the contents of 1 vial of Pseudomonas CN Supplement (SR0102) rehydrated as directed. Mix well and pour into sterile Petri dishes.

To Prepare Pseudomonas CFC Agar:
To 500ml of agar base cooled to 50°C add the contents of 1 vial of Pseudomonas CFC Supplement (SR0103) rehydrated as directed. Mix well and pour into sterile Petri dishes.

Precautions
Fresh media should be prepared as required. Molten agar should not be kept longer than 4 hours. Medium should not be stored and remelted. If swarming colonies of Proteus species are a problem in food samples then the incubation temperature can be lowered to 20°C for a period of 3-5 days. Chilled foods may carry a wide range of pseudomonads and the colonies on CFC Medium, incubated at lower temperatures, may be Pseudomonas fluorescens or Pseudomonas putida as well as Pseudomonas aeruginosa. Aeromonas species will also appear as pink/brown colonies, particularly from fish products.

Suspend 52g of MacConkey agar powder in 1 litre of distilled water. Bring to the boil to dissolve completely. Sterilise by autoclaving at 121°C for 15 minutes.

Storage conditions and Shelf life of MacConkey Agar (Oxoid)
Store the dehydrated MacConkey agar powder at 10-30°C and use before the expiry date on the label.
Store the prepared MacConkey agar at 2-8°C.
