Listeria Selective Enrichment Medium Preparation:
Reconstitute one vial as directed, and add the contents to 500ml of  Listeria Enrichment Broth Base (CM0862). Sterilize by autoclaving at 121°C for 15 minutes. Cool to 50°C and mix well before distributing into sterile containers in volumes as required.

Storage conditions and Shelf life
Listeria Selective Enrichment Supplement SR0141 as supplied should be stored at 2-8°C. When stored as directed the reagents are stable until the expiry date printed on the label.

Precautions
Listeria Selective Enrichment Supplement contains cycloheximide and is toxic if swallowed, inhaled or by skin contact. As a precaution when handling wear gloves and eye/face protection.
Acriflavine is activated by light and may form compounds inhibitory for Listeria.

EC Broth (Reduced Bile Salts), Oxoid Preparation:

Baird-Parker Agar Base (RPF), Oxoid Preparation:
Suspend 6.3g of Baird-Parker Agar Base (RPF) in 90ml of distilled water. Bring to the boil to dissolve completely. Sterilise by autoclaving at 121ºC for 15 minutes. Cool to 48ºC and add 1 vial of SR0122A, reconstituted as directed. Mix well and pour plates.

Prepared plates of medium are best used freshly prepared.

Precautions
Colonies of some contaminating organisms growing in close proximity to the coagulase positive colonies may partially digest the coagulase halo reaction.

Rogosa Agar, Oxoid Preparation:
Suspend 82g in 1 litre of distilled water and bring to the boil to dissolve completely. Add 1.32ml glacial acetic acid and mix thoroughly. Heat to 90-100°C for 2-3 minutes with frequent agitation. Distribute into sterile tubes, Petri dishes or bottles.

Vogel-Johnson Agar, Oxoid Preparation:
Suspend 61g in 1 litre of distilled water and bring gently to the boil to dissolve completely. Sterilise by autoclaving at 121°C for 15 minutes. Cool to 50°C and add 5.7ml of sterile 3.5% potassium tellurite solution SR0030 (equivalent to 20ml of 1% potassium tellurite).

Precautions

Czapek Dox Agar Modified, Oxoid Preparation:
Suspend 45.4g in 1 litre of distilled water. Bring to the boil to dissolve completely. Sterilise by autoclaving at 121°C for 15 minutes. Mix well before pouring. If required, adjust the reaction to pH 3.5 ± 0.2 by adding 10ml of Lactic Acid 10% (SR0021) per litre after sterilisation.

Chromogenic Salmonella Agar, Oxoid Preparation:
Suspend 25g in 500ml of distilled water and add the contents of one vial of Salmonella Selective Supplement (SR0194) reconstituted as directed. Mix well and bring to the boil with frequent agitation. DO NOT AUTOCLAVE. DO NOT HOLD AT BOILING TEMPERATURE. Cool to 50°C, mix well and pour into sterile Petri dishes.

Brilliance Salmonella Agar, Oxoid Preparation:
Suspend 27g of Brilliance Salmonella Agar Base in 500ml of distilled water. Add the contents of 1 vial of Salmonella Selective Supplement, resuspended as directed. Mix well and sterilise by bringing to the boil with frequent agitation. Cool to around 50ºC, mix well and pour into sterile Petri dishes.
NOTE: It is critical that the selective supplement is added prior to heating. When preparing volumes greater than 1 litre contact Oxoid Technical Support for directions.

Salmonella Selective Supplement should be stored in the dark at 2-8ºC. When stored as directed, the un-opened products will remain stable until the expiry date printed on the packaging.

Membrane Endo Agar LES, Oxoid Preparation:
Suspend 45 grams in 1 litre of distilled water. Add 12ml of a 10% w/v solution of Basic Fuchsin. Heat gently with frequent agitation until the medium boils. DO NOT AUTOCLAVE. Cool to 45°C and dispense into 50-60 mm dishes in 4 ml volumes. For larger dishes use sufficient medium to give an equivalent depth (approx. 1.5 mm).
Plates should be protected from light and may be stored for up to two weeks in the refrigerator.

Agar Bacteriological (Agar No.1) 500g, Oxoid

Agar Bacteriological (Agar No. 1, Oxoid) is a clear purified high gel strength bacteriological agar compatible with all culture media. This agar has low calcium (Ca) and magnesium (Mg) levels and has very high working gel strength (1.5% w/v).

Agar Bacteriological, Oxoid is compatible with all culture media. It can be formulated to either broth or agar of the same medium with very similar metal values. This characteristic is especially valuable in antimicrobial MIC studies where differences in mineral/metal content can profoundly influence the results. Due to the low mineral/metal content in Agar Bacteriological, Oxoid, it allows free diffusion of antimicrobial substances. As a result, it is a highly satisfactory agar for antimicrobial diffusion studies (disc diffusion susceptibility tests).

Pseudomonas Cetrimide Agar (Usp-Ep) 500g

Enumerate heterotrophic bacteria from drinking water samples with Thermo Scientific™ Oxoid™ R2A Agar (Dehydrated). This nutritionally reduced medium was specially designed to enhance the recovery of stressed and chlorine damaged bacteria from treated water samples. Stressed or injured organisms during water treatment are unable to grow on high nutrient media since faster-growing organisms outgrow the former. Therefore the use of a low nutrient medium like R2A Agar incubated for longer incubation periods allows these stressed organisms to grow well.

Urea Agar Preparation:
Suspend 2.4g in 95ml of distilled water. Bring to the boil to dissolve completely. Sterilise by autoclaving at 115°C for 20 minutes. Cool to 50°C and aseptically introduce 5ml of sterile 40% Urea Solution SR0020. Mix well, distribute 10ml amounts into sterile containers and allow to set in the slope position.

Precautions
The alkaline reaction produced in this medium after prolonged incubation may not be caused by urease activity. Check using medium without urea.
Do not heat or reheat the medium because urea decomposes very easily.
For the detection of urease-positive Proteae the reaction must be read within the first 2-5 hours of incubation.

CLED Medium with Andrade Indicator 500g
