Chromogenic Salmonella Agar, Oxoid Preparation:
Suspend 25g in 500ml of distilled water and add the contents of one vial of Salmonella Selective Supplement (SR0194) reconstituted as directed. Mix well and bring to the boil with frequent agitation. DO NOT AUTOCLAVE. DO NOT HOLD AT BOILING TEMPERATURE. Cool to 50°C, mix well and pour into sterile Petri dishes.

CR-SMAC Agar, Oxoid Preparation:
Suspend 28.25g in 500ml of distilled water. Mix well and sterilise by autoclaving at 121°C for 15 minutes. Cool to approximately 50°C and aseptically add the contents of one vial of Cefixime Supplement SR0191, reconstituted as directed. Mix well and pour into sterile Petri dishes.
To reconstitute Cefixime Supplement, aseptically add 2ml sterile distilled water to 1 vial of supplement. Mix gently to dissolve.

Other enteric organisms e.g. some Shigella species are also glucuronidase positive.

VRBL Agar (ISO) should be stored in the tightly capped original container at 10-30°C. When stored as directed, the un-opened product will remain stable until the expiry date printed on the packaging.
Locally prepared VRBL Agar (ISO) plates can be stored for up to 2 weeks when made from CM0968 according to the manufacturer’s instructions and stored at 2-8°C, out of direct sunlight. A longer shelf life may be attainable, but should be validated under the relevant, local manufacturing and storage conditions.
Precautions
VRBL Agar (ISO) should be prepared, cooled to 44-47°C and used within 4 hours.
VRBL Agar (ISO) is for laboratory use only. Do not use the dehydrated medium beyond the stated expiry date or if the product shows any sign of deterioration.

Anaerobe Basal Broth, Oxoid Preparation:
Suspend 35.4g of Anaerobe Basal Broth in 1 litre of distilled water. Bring to the boil to dissolve. Distribute into final containers. Sterilise by autoclaving at 121°C for 15 minutes.
Storage conditions and Shelf life
Anaerobe Basal Broth should be stored tightly capped in the original container at 10-30°C. When stored as directed, the medium will remain stable until the expiry date printed on the label.

Brilliance E. coli/Coliform Agar, Oxoid Preparation:
Suspend 55.8g of Brilliance E.coli/coliform agar in 1 litre of distilled water. Sterilise by autoclaving at 121°C for 15 minutes. Cool to approximately 50°C. Mix well and pour and into sterile Petri dishes.
Storage conditions and Shelf life
Store the dehydrated medium at 10-30°C, and use before the expiry date on the label.
Store the prepared medium at 2-8°C.

Some pathogenic strains, typically Escherichia coli O157:H7 are usually glucuronidase negative and therefore will not give purple colonies on this medium.

Detect and enumerate Escherichia coli in food and animal feed samples with Thermo Scientific™ Oxoid™ Typtone Bile X-Glucuronide (TBX) Medium (Dehydrated). The medium meets the formulation and performance criterion laid down in ISO 16649 and ISO 11133:2014.
TBX Medium builds on the speed, reliability and recovery from frozen samples and detection of poor lactose fermenters offered by the original Tryptone Bile Agar, with the addition of a chromogenic agent, X-glucuronide, which detects glucuronidase activity, an enzyme shown to be highly specific for Escherichia coli.

Campylobacter Agar (Karmali), Oxoid Preparation:
Add 21.5g of Campylobacter Agar Base (Karmali) to 500ml of distilled water and bring to the boil to dissolve. Sterilise by autoclaving at 121°C for 15 minutes. Cool to 50°C. Aseptically add 1 vial of Campylobacter Selective Supplement (Karmali) SR0167 or Modified Karmali Selective Supplement SR0205, reconstituted as directed.. Mix well and pour into sterile Petri dishes.

Campylobacter Agar Base (Karmali) should be stored tightly capped in the original container in a cool, dry place away from bright light. When stored at 10-30°C the medium will remain stable until the expiry date printed on the label.

Selectively enumerate Bacillus cereus in food samples with Thermo Scientific™ Oxoid™ MYP Agar (Dehydrated). The medium relies upon mannitol fermentation and phospholipase C production to differentiate Bacillus cereus from other members of the species.
Mannitol-negative Bacillus cereus form pink colored colonies due to the presence of Phenol Red Indicator. Egg yolk emulsion helps in the differentiation of phospholipase C producing colonies, which are surrounded by a zone of the white precipitate. Polymixin B Sulphate acts as the inhibitor to restrict the growth of Gram-negative bacteria.

MYP Agar, Oxoid Preparation:
Suspend 21.5g in 450ml distilled water and bring gently to the boil to dissolve. Sterilise by autoclaving at 121°C for 15 minutes.
Cool to approximately 49°C and aseptically add 50ml Egg Yolk Emulsion SR0047 and 1 vial of Polymyxin B Supplement SR0099, reconstituted as directed. Mix well and pour into sterile petri dishes.

Yeast and Mold Agar, Oxoid Preparation:
Suspend 41.0g in 1 litre of distilled water and bring to the boil to dissolve. Sterilise by autoclaving at 121°C for 15 minutes. The medium may be rendered selective after sterilisation by acidifying to pH 4.0 with 12-15ml of Lactic Acid SR0021 after cooling to 50°C. Do not reheat after making this addition. Mix well and pour into sterile Petri dishes.

The prepared medium may be stored for up to 2 weeks at 2-8°C.
